I recently purchased the Domain Name for my site. Got it from Register.com, and transfered the DNS over to my Hosting Site. My question here is..... Do I need to reinstall PostNuke and all the Modules I have, so that it resets to the new address? www.mydomain.com? Or will this just happen automatically as the DNS changes over and points to the sites already installed location?

If I understand your message correctly then no you should not have to do anything about reinstalling your site. It will just take a few days, I believe they normally indicate three days, before the information is set to point to the new address of a site. So it may take a few days before your site is visible to the world through the domain name but you should still be able to access it through a temporary address your hosting provider gave you when you set your account up with them.

Yeah, it did. It hit today. I was just making sure I wouldn't have to do anything further. I have a few bugs now that I'm tracking down and trying to fix. Didn't wish to have to start all over. Thanks for your response.
